They'd have to keep it to the five main members. Fuck Aquaman and Martian Manhunter. Five seems to be the limit before you make a movie collapse under the weight of too many things going on. (and another reason that the FF shitfests are such a pity). Use Supes, Bats, WW, GL, and Flash. And that's IT.
And I'd say copy the Marvel blueprint in terms of tone. It worked for Avengers. Steal that shit! If they can pull off Thor, Iron Man, Cap and Hulk, they sure as hell COULD do the JLA.
They won't mind you, but it could be done.

Sure they could. If you squint a bit, we basically could have been watching Superman, Batman, Wonder Woman, Green Arrow, Black Canary, and Green Lantern fight Parademons popping out of Boom Tubes sent by Darkseid, anyway.

Another reason why I know it COULD work!

Maybe it's a bias, but JSA would look less cheesy on film in my book. Sandman, Doc Fate, Atom, Canary, Mid-Nite, fuck...even Johnny T. could work with a CGI T-Bolt as comic relief.

No, you're right. More muted colors and less over the top characters would probably work better.

Late Forties backdrop would set it apart too.

I kind of think this would have to be done with the "unpowered" cast members only. Give a pulpy flavor.
Sandman, Starman, Hourman, Dr. Mid-Nite, etc would make it a sort of ensemble cast that could rock.

So Wonder Woman is the Hulk? "Puny god."

Meanwhile even the best of DC (the Nolan bat-verse) always seems ashamed by the fact they're comic movies. They get grounded in realism. Everything is re-imagined.
While I can't argue with the success of Nolan's Batman projects -- and admit that I enjoyed them for what they were, I think they were a little too grounded in reality. A little escapism is good.

So Wonder Woman is the Hulk? "Puny god."
It's not an exact fit... I was actually thinking WW was a parallel for Thor, being the mythological character. But Supes is definitely the alien powerhouse game-changer, like the Asgardian. And Batman is sort of split between being a Cap and Iron Man type. But you get the gist.

It all comes down to this: you get the right screenwriter and the right director -- then anything is possible. Well, a good casting director too, but you get the idea.
Trust me, that's what it comes down to, far more than budgets or corporate meddling.

A billion dollars in less than three weeks for the Avengers. The misery piles on for Warner's.
